Perhaps your tiff version is monochrome or a different resolution? If you want to go the DNG route I might recommend either scanning to tiff, do your spotting and basic corrections on it, and then convert to DNG, or just use ACR directly on the tiff. Not sure if Lightroom can be made to work on tiffs.
