Ken I don't know which applications/methods/hardware that you are using
apart from your RIP. With your OS it gets more complicated too if you have
moved to 64 bit, which is preferable for Photoshop too.
Photoshop CS5 partially breaks the ability to develop printing profiles due
to a change in the way Colour adjustment is selectable.
You can still use existing profiles however you cannot produce new ones
using the existing method (which involves selecting No Color Adjustment in
the printing of the targets to be analysed).
Adobe state that method did not work for every application. I had no problem
under CS2,3,4.
Adobe says that they are developing a  free tool that can be used to make
printing profiles instead of the existing methods.
You can find more detail on this in the Adobe forums if desired.
