Scanning with DI on at 5400 dpi takes almost exactly twice as long for me (approximately six minutes vs. three per frame). There is an unavoidable slight decrease in sharpness. This can be good too as it softens the "grain" a bit. (I know that technically XP2 doesn't have "grain".) But this loss of sharpness can be recovered in Photoshop and the time saved in not having to remove dust spots one at time in PS is definitely worth the longer scan times.
